Abstract Herein we report the use of ultrasonic irradiation (US) in the synthesis of six new semi-synthetic selenium-containing chrysin derivatives by a simple and effective methodology utilizing CuI as catalyst, in good to excellent yields (60–89%). It was observed that US accelerates the reaction compared to conventional heating with excellent selectivity for diselenylated products. Compounds were tested for their antioxidant and anticancer activities in vitro and it was observed that the presence of selenium in the A-ring of chrysin enhanced both antioxidant and anticancer properties. Semi-synthetic 6,8-bis( o -tolylselanyl)-chrysin 3b has the best radical scavenging activity of DPPH (I max : 39.79 µM) and ABTS + (IC 50 : 6.5 µM) radicals. Similarly, in the Reactive Species (RS) assay, 3b showed high antioxidant activity in mice cortex (IC 50 : 5.67 µM), whereas 6,8-bis( p -anisoylselanyl)-chrysin 3c was the more active in the hippocampus (IC 50 : 5.63 µM). The Se-chrysins were effective in prevention of lipid peroxidation, highlighting 6,8-bis( p -fluorophenylselanyl)-chrysin 3d in cortex (IC 50 : 0.54 µM) and 3b in hippocampus (IC 50 : 0.27 µM). In addition, 3d was effective in inhibiting human lung adenocarcinoma (A549) cells growth, with a IC 50 of 19.9 µM after 72 h of treatment, while 6,8-bis( p -anisoylselanyl)-chrysin 3c presented the higher antiproliferative activity after 48 h of treatment (IC 50 of 41.4 µM).

Abstract Under physiological conditions, cells are in a stable state known as redox homeostasis, which is maintained by the balance between continuous ROS/RNS generation and several mechanisms involved in antioxidant activity. ROS overproduction results in alterations in the redox homeostasis that promote oxidative damage to major components of the cell, including the biomembrane phospholipids. Lipid peroxidation subsequently generates a diverse set of products, including α,β-unsaturated aldehyd...

The use of selenium-containing heterocyclic compounds as potent cancer chemopreventive and chemotherapeutic agents has been well documented by a large number of clinical studies. In this study we developed a new approach to synthesize four benzimidazole-containing selenadiazole derivatives (BSeDs). The method uses a combination of peptide coupling reagents and microwave irradiation. This strategy features milder reaction conditions, higher yields, and shorter reaction times. The synthetic BSeDs ...

Abstract Redox biology has evolved from studies of the pathology that involves oxidants to an understanding of how oxidants participate in normal as well as aberrant signal transduction. Although the concept that signal transduction involved changes in the redox state dates from the 1930s, the modern history of redox biology began with the discovery of superoxide dismutase by McCord and Fridovich. The initial focus was on free radicals and damage of macromolecules, which remains an important top...

Abstract Chrysin, one of natural flavonoid compounds, has recently been found to possess anti-inflammatory, antiallergic and anticancer properties. To increase its anticancer effects, 5 chrysin derivates were synthesized on the base of DNA intercalator structure. The inhibiting effects of chrysin and its derivatives on cancer cells Hela, BGC823, MCF-7, HepG2, and normal cells HEK-293, were evaluated by MTT assays. 5-(2′-amino) phenyl-7-cyclohexanemethylchrysin (Ch-1), a unique chrysin derivate, ...

Objectives: Chrysin, an active natural bioflavonoid found in honey and many plant extracts, was first known for its antioxidant and anti-inflammatory effects. The fact that antioxidants have several inhibitory effects against different diseases, such as cancer, led to search for food rich in antioxidants. In this study, we investigated the antiproliferative and apoptotic effects of chrysin on the cultured human breast cancer cells (MCF-7). Materials and Methods: Cells were cultured in Roswell Pa...

Chrysin is a major active ingredient of flavonoids, known to exhibit protective effects against various types of cancer. However, the anticancer role of chrysin against hepatocellular carcinoma (HCC) and the underlying molecular mechanisms remain unclear. In order to evaluate the effects of chrysin on cell viability and apoptosis in human HCC, HepG2 and QGY7701 cells were used in the present study. Cell viability was monitored using an MTT assay. In addition, an Annexin V-fluorescein isothiocyan...

Abstract The development of new drugs to treat painful and inflammatory clinical conditions continues to be of great interest. The present study evaluated the antinociceptive and anti-inflammatory effects of 4-phenylselenyl-7-chloroquinoline (4-PSQ). Mice were orally (p.o.) pretreated with 4-PSQ (0.1–25 mg/kg), meloxicam (25 mg/kg, a reference drug) or vehicle, 30 min prior to the acetic acid, formalin, hot-plate and open-field tests. 4-PSQ reduced abdominal writhing induced by acetic acid and i...

CombretastatinA-4 (CA-4, 1a, Fig. 1), a natural stilbene isolated from the South African tree Combretum caffrum, is a powerful inhibitor of tubulin polymerization that displays cytotoxic and antivascular activities by binding at the colchicine site of tubulin1. CA-4P (1b), the water-soluble pro-drug of CA-4, has shown promising results in human cancer clinical trials2. The structure-activity relationship (SAR) study of CA-4 indicated that the presence of a cis-double bond and 3,4,5-trimethoxyphe...

Abstract Depression is one of the most commonly diagnosed neuropsychiatric disorders and several studies have demonstrated a role for selenium in mood disorders. For this reason, the present study investigated the role of the monoaminergic system in the antidepressant-like action of (octylseleno)-xylofuranoside (OSX), an organoselenium compound, in the tail suspension test (TST) in mice. For this purpose, OSX (0.001–10 mg/kg) was administered orally (p.o.) 30 min prior to testing, and all of the...

Abstract Inflammatory bowel disease (IBD) is a chronic disease that occurs in the intestinal tract. Phyto-ingredients have been evaluated for their ability to protect against IBD because of their anti-inflammatory activities. In our previous study, we identified a novel derivative of chrysin (HE-chrysin) using irradiation technology, which exhibited stronger anti-cancer activity in human colorectal cancer cells than the original chrysin. Here, to determine whether HE-chrysin is a new therapeutic...

Transition metal catalysed C–H functionalization has reached an exciting level of sophistication, and, today, it represents a paradigm shift from the standard logic of synthetic chemistry. The direct conversion of C–H bonds into C–heteroatoms remains, however, a critical challenge. Nowadays, there is a great demand in general synthetic chemistry in, for example, the materials science for the development of straightforward C–Se bond formation, in order to fulfil the practical requirements. In thi...
