Abstract
The analytical chemistry of lipid peroxidation is reviewed with special attention to literature that has appeared in the last four years. The pathways of the cascade of reactions are first described and current analytical practice discussed. Selected assays, classified by analyte, are then considered as indices of lipid peroxidation; these mainly involve ultraviolet and visible spectrophotometry, fluorescence and chemiluminescence.

A new method for the convenient and sensitive detection of aldehydes and ketones in environmental samples was developed. This method is described in detail for the measurement of carbonyls in water samples, but it is also potentially applicable to their determination in air. Derivatization of carbonyls is performed using N-(5-dimethylamino-1-naphtalenesulphonamido)-3-oxapentane-1,5-dioxyamine (dansyloxyamine, DNSOA). The resulting oxime ethers are separated by using reversed phase liquid chromat...

An HPLC (high performance liquid chromatography) method with laser induced fluorescence (LIF) detection is described for the determination of 4-hydroxy-2-nonenal (HNE) formed from lipid peroxidation in rat hepatocytes. Carbonyl compounds were fluorescently labelled by incubating the hepatocyte samples with a tagging reagent, 4-(2-carbazoylpyrrolidin-1-yl)-7-nitro-2,1,3-benzoxadiazole (NBD-ProCZ), at 60°C for 10 min. The hydrazone derivatives were extracted with a C18 solid phase extraction (SPE)...

A high-performance liquid chromatographic (HPLC) method is described for the quantitative determination of some aliphatic aldehydes and β-hydroxyaldehydes as their 2,4-dinitrophenylhydrazone derivatives. A method is described for the preparation of derivatives for those β-hydroxyaldehydes where no reference compounds of known purity are available. The detection limit of the method was 4.3−21.0 μg/L, depending on the aldehyde.

An important emerging issue in chemical carcinogenesis is the role that products of endogenous metabolism play in formation of covalently modified DNA. One example is the formation of α,β-unsaturated aldehydes as a result of endogenous and drug-stimulated lipid peroxidation. Malondialdehyde (MDA), crotonaldehyde (CR), 2-hexenal (HX), and 4-hydroxy-2-nonenal (HNE) react covalently with 2′-deoxyguanosine (dG) and 2′-deoxyadenosine (dA) residues on DNA to form promutagenic cyclic adducts that may b...

A simple HPLC method for the determination of malondialdehyde (MDA) in oxidised vegetable oils was developed and the results were compared with those given by a spectrophotometric method. Vegetable oil was steam-distilled in a Kjeldahl distillation apparatus and the MDA was determined in the aqueous distillates by HPLC, using a µ-Bondapak C18 column, with a mobile phase of 1% acetic acid–acetonitrile (85 + 15, v/v). A total time of 2 min was necessary to assay each distillate and only MDA was de...

The oxidative chemiluminescence of phenylhydrazines can be applied to the determination of carbonyl compounds by utilising the attenuation of the signal which occurs on conversion to the corresponding phenylhydrazone. Using an assay for 2,4-dinitrophenylhydrazine, off-line derivatisation for a period of 2 h was found to be optimised for a temperature of 30 °C and for a sulfuric acid concentration of 0.05 M. The optimum conditions for 2,4-dinitrophenylhydrazone formation can be understood in term...

Lipid oxidation is a significant problem relative to off-flavour and off-odour, and warmed-over flavour appears to be related to lipid oxidation in meat. In this review article the thiobarbituric acid (TBA) test for lipid oxidation measurements is described. Works on other techniques to measure lipid oxidation, as determination of malondialdehyde content by high performance liquid chromatography or gas chromatography are reviewed. A detailed description of the different TBA test procedures, poss...

Abstract A method for in vivo evaluation of lipid peroxidation in the extracellular space of anaesthetized rat brain cortex was developed. This method involved the use of microdialysis perfusion and high-performance liquid chromatography. The microdialysates, eluted from implanted probes, were reacted with thiobarbituric acid (TBA) prior to analysis by an HPLC system equipped with a fluorescence detector (excitation and emission wavelengths were 515 and 550 nm, respectively). Lipid peroxidation ...

Abstract A chemiluminescence(CL)-high performance liquid chromatography (HPLC) system, where CL detector was equipped with normal phase HPLC, was used in order to specifically detect lipid hydroperoxide in the heart of doxorubicin (DXR) administered mice. Luminol and cytochrome c were used as the luminescence reagents specific for the hydroperoxide. Phosphatidylcholine hydroperoxide was detected in the heart, indicating participation of lipid peroxidation in the DXR-induced cardiotoxicity. This ...

BACKGROUND: Bentonite is natural clay composed mainly of montmorillonite with others associated minerals such as feldspar, calcite and quartz. Owing to its high cation exchange, large surface area and ability to form thixotropic gels with water and to absorb large quantities of gas, it presents a large medicinal application. OBJECTIVE: This review focuses on promising potential of bentonite clays for biomaterial design and for therapeutic purposes. METHODS: PubMed, ACS publications and Elsevier ...

Abstract Free radical scavengers like α-phenyl- N - tert -butylnitrone (PBN) and 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox) have been widely used as protective agents in various biomimetic and biological models. A series of three amphiphilic Trolox and PBN derivatives have been designed by adding to those molecules a perfluorinated chain as well as a sugar group in order to render them amphiphilic. In this work, we have studied the interactions between these derivatives and ...

Abstract Polyunsaturated fatty acids are particularly sensitive to the damages due to reactive oxygen species and lipid oxidation has been reported to be involved in the degradation of food as well as in the early stages of several diseases. Our objective was to study the mechanisms of action of flax (Linum usitatissimum) phenolic compounds to prevent membrane lipid oxidation. To do so, several biophysical techniques (oxidative stress, surface tension, fluorescence spectroscopy and HPLC) were us...

Abstract In this contribution an electrochemical study is described for the first time of lipid peroxidation and the role of antioxidant on lipid protection using large unilamellar vesicles (LUVs). In order to simulate the cell membrane, LUVs composed of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) were used. A vesicle-modified electrode was constructed by immobilizing DOPC LUVs onto carbon paste electrodes (CPEs). Lipid peroxidation was studied electrochemically by incubating the vesicle-mod...

Abstract Okadaic Acid (OA) is a marine toxin responsible for DSP (Diarrheic Shellfish Poisoning) in humans produced by dinoflagellate. The genotoxic and cytotoxic effects of OA have been well reported in mammalian experimental animals and in vitro cultured cells. However, there are no available investigations regarding the involvement of the oxidative stress pathways in OA toxicity, especially on aquatic animals such as fish. In this context, we aimed in the present work to demonstrate whether O...

Abstract Free radical scavengers such as α-phenyl- N-tert- butylnitrone (PBN) have been widely used as protective agents in several biological models. We recently designed two PBN derivatives by adding a cholesterol moiety to the parent nitrone to increase its lipophilicity. In addition to the cholesterol, a sugar group was also grafted to enhance the hydrophilic properties at the same time. In the present work we report on the synthesis of a third derivative bearing only a cholesterol moiety an...
