Adverse drug reactions (ADRs) remain a major complication of drug therapy and can be classified as ‘on-target’ or ‘off-target’ (idiosyncratic) reactions. On-target reactions can be predicted from the known primary or secondary pharmacology of the drug and often represent an exaggeration of the pharmacological effect of the drug.
