High Diversity of Ligularia dictyoneura in Chemical Composition and DNA Sequence

The chemical composition of root extracts of the title species collected at 20 different places in the Hengduan Mountains of China was examined. From these samples, a total of 17 eremophilane derivatives were isolated, three of which were new franoeremophilane derivatives: 3 beta-acetoxy-6 beta-(angeloyloxy)furanoeremophilan-10 beta-ol, 1 alpha-acetoxyfuranoeremophilan-15,6 alpha-olide, and 6 beta-[2-(hydroxymethyl)prop-2-e noyloxy] furanoeremophil-1(10)-ene. Based on the chemical composition, the samples could be classified into as many as seven types: one type containing non-furanoeremophilane derivatives and the other six containing furanoeremophilane derivatives with different oxidation levels. Results of DNA sequencing of the atpB-rbcL region and the internal transcribed spacers of the ribosomal RNA gene also indicated a high diversity in the species.

The chemical composition of a sample of the title species collected in Pianma, Lushui County, Yunnan Province of China, was examined. Two 3,6,10 oxygenated and two 3,6-oxygenated furanoeremophilanes were isolated. Structures of two new compounds were elucidated. The 3,6-oxygenated furanoeremophilanes were not detected in a sample of the same species previously collected in Laojunshan, Jianchuan County, Yunnan, indicating chemical diversity in the species.

Ligularia speices are widely used in Asian folk medicines for the treatment of various human diseases. Eremophilane-type sesquiterpenes are abundant and typical secondary metabolites found in this genus. Over 500 eremophilanes reported from members of Ligularia are reviewed in this article together with bioactivity data in an effort to highlight the development in this field.

Five new 12-oxygenated eremophilane sesquiterpenoids bearing a B-ring enone system were isolated from Ligularia lingiana S.W. Liu (Asteraceae) collected in China. Compound 5 was a Diels-Alder adduct of 3 alpha-angeloyloxy-7 beta H-8-oxoeremophila-9,11(13)-dien-12-al and the monoterpene, ocimene.
