Diversity of Ligularia kanaitzensis in sesquiterpenoid composition and neutral DNA sequences

Abstract
Twenty-six eremophilane-type sesquiterpenoids, including six new compounds, were isolated from the title species. One of the new compounds, kanaitzensol, suggested the presence of an enzyme for the conversion of eremophil-7(11)-en-8-one derivative to furanoeremophilanes. The plant was collected at 15 locations in Yunnan Province of China and found to be diverse with respect to its sesquiterpenoid composition. DNA sequencing also revealed a large intraspecific diversity of the plant. (c) 2008 Elsevier Ltd. All rights reserved.

Intra-specific diversity in Liularia vellerea growing in the northwestern to central Yunnan province of China was studied by chemical and genetic approaches. The chemical constituents of the root of Ligularia duciformis, collected in Yunnan Province of China, was studied and two new oplopane-type sesquiterpenoids were isolated, whose structures were determined on the basis of spectroscopic evidence. Quite recently, oplopane-type compounds were found in Ligularia narynensis, and this is the second example to isolate oplopane-type sesquiterpenoids from Ligularia plants.

Four samples of Ligularia virgaurea collected in China afforded three new eremophilane sesquiterpenoids, one new norsesquiterpenoid, and two new dinorsesquiterpenoids as well as 28 known compounds. The chemical composition of a sample of the title species collected in Pianma, Lushui County, Yunnan Province of China, was examined. Two 3,6,10 oxygenated and two 3,6-oxygenated furanoeremophilanes were isolated. Structures of two new compounds were elucidated. The 3,6-oxygenated furanoeremophilanes were not detected in a sample of the same species previously collected in Laojunshan, Jianchuan County, Yunnan, indicating chemical diversity in the species.

Ligularia speices are widely used in Asian folk medicines for the treatment of various human diseases. Eremophilane-type sesquiterpenes are abundant and typical secondary metabolites found in this genus. Over 500 eremophilanes reported from members of Ligularia are reviewed in this article together with bioactivity data in an effort to highlight the development in this field.
