Chemical constituents of Ligularia virgaurea and its diversity in southwestern Sichuan of China

Chemical constituents of root extract and the nucleotide sequences of the atpB-rbcL intergenic region and the internal transcribed spacers (ITSs) of the ribosomal RNA gene were studied for Ligularia virgaurea var. virgaurea collected in southwestern Sichuan Province of China. Eleven samples were collected. Four of them were found to contain four new furanoeremophilanes, virgaurenones A-D, as well as a new cremophilanolide, virgaurenolide A. The other samples contained different furanoeremophilanes and their derivatives including nor-type of compounds, two of which were new. Diversity was found to be present in the nucleotide sequences as well. The chemical composition was found to be correlated with the ITS variation but not with the geographic distribution of the samples. (c) 2006 Elsevier Ltd. All rights reserved.

Intra-specific diversity of Ligularia pleurocaulis was examined from both chemical and genetic aspects, using furanoeremophilanes as the chemical index and the nucleotide sequences in the atpB-rbcL and the ITS1 regions as the genetic index. Three Ligularia species (L. tongolensis, L. cymbulifera, and L. atroviolacea) were examined with respect to the chemical composition and nucleotide sequence. Seven eremophilane-type sesquiterpenes have been isolated from Petasites japonicus ssp. giganteus collected in Hokkaido, and two of them have been found to be new. Eremopetasitenin A 3 was a lactone containing an epoxide at the C-7 and C-8 positions, whose structure was established mainly based on spectroscopic evidence including 2D NMR spectra. Two new norsesquiterpenes were isolated from the extracts of the roots and rhizomes of Ligularia lapathifolia (Franch.) Hand.-Mazz. Their structures were identified as 2-acetyl-3a-methyl-5-(2-methyl-but-2-enoyloxy)3a, 4,5,6,7,7a-hexahydro-1H-indene-4-carboxylic acid (1) and 2-acetyl-8a-methyl-2-(2-methyl-but-2-enoyloxy)6- oxo-1,2,3,4,4a, 5,6,8a-octahydro-naphthalene-1-carboxylic acid (2), respectively, on the basis of spectral data and for 1 by single-crystal X-ray analysis.

Abstract Four sesquiterpenoids were isolated from Petasites japonicus (Siebold and Zucc.) Maxim. ssp. giganteus (F. Schmidt ex Trautv.) Kitam. (Compositae, Japanese name: Akitabuki) as allelochemicals. Their structures were elucidated on the basis of spectroscopic methods and their allelopathic effects were assessed.

Ligularia speices are widely used in Asian folk medicines for the treatment of various human diseases. Eremophilane-type sesquiterpenes are abundant and typical secondary metabolites found in this genus. Over 500 eremophilanes reported from members of Ligularia are reviewed in this article together with bioactivity data in an effort to highlight the development in this field.

Further chemical investigation of two Ligularia virgaurea samples collected in China resulted in the isolation of 21 new compounds, two of which were bakkane-type sesquiterpenoids bearing an anhydride-type ring C, which was a previously unknown partial structure. An extract was prepared by refluxing with ethanol and steam distillation, and its toxic effect was tested in vitro against Sarcoptes scabiei.
