Although icosahedral viruses have obvious and highly symmetrical features, asymmetric structural elements are also present. Asymmetric features may be inherent since the genome and location of minor capsid proteins are typically incorporated without adhering to icosahedral symmetry. Asymmetry also develops during the virus life cycle in order to accomplish key functions such as genome packaging, release, and organization. However, resolving asymmetric features complicates image processing during...

Abstract Two novel tandem catalysis approaches for the chiral synthesis of S -alcohols from reduction of their prochiral ketones with Horse Liver Alcohol Dehydrogenase (HLADH), and selective C-H oxidation reactions with protein engineered Cytochrome P450s, are presented. We utilized a co-factor regeneration procedure with three biomimetic NAD + models that do not contain the pyrophosphate, nor the adenosine group, and either/or a ribose, N-1-benzylnicotinamide triflate, 1 , N-4-methoxybenzylnico...

Glutamate-1-semialdehyde-2,1-aminomutase (GSAM) catalyzes the isomerization of glutamate-1-semialdehyde (GSA) to 5-aminolevulinate (ALA) and is distributed in archaea, most bacteria and plants. Although structures of GSAM from archaea and bacteria have been resolved, a GSAM structure from a higher plant is not available, preventing further structure–function analysis. Here, the structure of GSAM from Arabidopsis thaliana (AtGSA1) obtained by X-ray crystallography is reported at 1.25 A resolution...

Abstract A novel ficin form, named ficin E, was purified from fig tree latex by a combination of cation-exchange chromatography on SP-Sepharose Fast Flow, Thiopropyl Sepharose 4B and fplc-gel filtration chromatography. The new ficin appeared not to be sensitive to thiol derivatization by a polyethylene glycol derivative, allowing its purification. The protease is homogeneous according to PAGE, SDS–PAGE, mass spectrometry, N-terminal micro-sequencing analyses and E-64 active site titration. N-ter...

Aspartate and glutamate are two key amino acids used in biosynthesis of many amino acids that play vital role in cellular metabolism. Aspartate aminotransferases (AspATs) are required for channelling nitrogen (N2) between Glu and Asp in all life forms. Biochemical and genetic characterization of AspATs have been lacking in N2-fixing cyanobacteria. In this report, five putative AspAT genes (alr1039, all2340, alr2765, all4327 and alr4853) were identified in the N2-fixing heterocystous cyanobacteri...

A plant protease named microcarpain was purified from the latex of Ficus microcarpa by acetonic (20–40 % saturation) precipitation, Sephadex G-75 filtration, and Mono Q-Sefinose FF chromatography. The protease was purified with a yield of 9.25 % and a purification factor of 8. The molecular weight of the microcarpain was estimated to be 20 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The purified enzyme showed maximum activity at pH 8.0 and at a temperature of 70 ...

Enzymes are the catalysts used by living organisms to accelerate chemical processes under physiological conditions. In this chapter, we illustrate the current view about the origin of their extraordinary rate enhancement based on molecular simulations and, in particular, on methods based on the combination of Quantum Mechanics and Molecular Mechanics potentials which provide a solution to treat the chemical reactivity of these large and complex molecular systems. Computational studies on Dihydro...
The microbial production of protease is the need of rapidly growing industrial field. So due to its wide utility protease production becomes the centre of attraction for commercial market. The present study emphasized on microbial production and optimization of protease enzyme. The potential protease production depends on effective strain selection, for this purpose we took different samples of stagnant water from different locations. Then we isolated the single colony of protease producing micr...
