Abstract A homogeneous extracellular polysaccharide of Lachnum YM261(LEPS-1) with a molecular weight of 21670 Da was characterised. According to HPGPC, IR, periodate oxidation and Smith degradation, GC–MS and 1 H NMR analysis, the results indicated that LEPS-1 was a glucan linked by the β-(1 → 3)- d -pyran glycosidic bond. The microwave method was used to extract Lachnum calyculiforme polysaccharides (LCP). By orthogonal experiments, the optimal conditions of extraction were the microwave temperature was 120 °C, extraction time was 20 min, ratio of water to raw material was 60:1, and the yield of polysaccharides was 12.56 %. The correlations between the yield of intracellular polysaccharide (LIP), the yield of extracellular polysaccharide (LEP) and the biomass of a Lachnum YM281 in submerged fermentation were studied. Results showed that there was a significant linear relationship between the yield of LIP (Y) and the biomass (Z): Z=12.561Y-0.0148 (R 2 =0.985).
