IFN-γ is an enigmatic cytokine that shows direct anti-viral effects, confers upregulation of MHC-II and other components relevant for antigen presentation, and that adjusts the composition and balance of complex cytokine responses. It is produced during immune responses by innate as well as adaptive immune cells and can critically affect the course and outcome of infectious diseases, autoimmunity, and cancer. To selectively analyze the function of innate immune cell-derived IFN-γ, we generated c...

Cytomegalovirus is a DNA-encoded β-herpesvirus that induces STING-dependent type 1 interferon responses in macrophages and uses myeloid cells as a vehicle for dissemination. Here we report that STING knockout mice are as resistant to murine cytomegalovirus (MCMV) infection as wild-type controls, whereas mice with a combined Toll-like receptor/RIG-I-like receptor/STING signaling deficiency do not mount type 1 interferon responses and succumb to the infection. Although STING alone is dispensable f...

Mature blood and immune cells show broad inter- and intra-cell type-specific heterogeneity with respect to metabolic activity, gene expression patterns, differentiation capacity and responsiveness to cytokines such as interferons (IFNs). This diversity is of crucial importance to maintain the capacity of the immune system to appropriately react to a wide range of pathogenic insults, thereby mediating protective immunity. However, it remains poorly understood how such heterogeneity is generated a...

Quiescent long-term hematopoietic stem cells (LT-HSCs) are efficiently activated by type I interferon (IFN-I). However, this effect remains poorly inv

Velten et al. use single-cell transcriptomics and functional data to map the early lineage commitment of human haematopoietic stem cells as a continuous process of cells passing through transitory states rather than demarcating discrete progenitors.

The cytomegalovirus (CMV), a member of the beta-herpesvirus family, is widely spread among the world population and causes lifelong latent infections, which can lead to severe diseases in immunocompromised patients. Therefore, it is important to understand the interplay between different immune mechanisms that are essential for the control of CMV infection. To dissect the role of different recognition platforms, MyD88Trif−/− and Cardif−/− mice devoid of TLR- or RLH-dependent signaling, respectiv...
