Airway inflammation (AI) and hyperresponsiveness (AHR) are two asthma hallmarks. However, the function of particular cell types, i.e. eosinophils or T-cells, in the development of AHR is still not unambigously elucidated. We hypothesized that challenge of mice with varying antigen doses could provide new insights into the complex mechanisms by which AI enhances AHR. AHR was detected only in mice challenged with larger OVA dose, indicating dependence of AHR on the antigen dose used for challenge....

A series of 3-keto and 3-O-acyl derivatives of both 6-O-alkyl-8a-aza-8a-homoerythromycin A and 6- O-alkyl-9a-aza-9a-homo-erythromycin A were synthesised and tested against gram-positive and gram-negative bacteria. Derivatives of 8a-aza-8a- homoerythromycin A have potent antibacterial activity against not only azithromycin-susceptible strains, but also efflux (M) and inducible macrolide-lincosamide-streptogramin (iMLSB) resistant Gram-positive pathogens, while the corresponding 9a-isomers were le...

Macrolide antibacterials inhibit the production of various cytokines and the migration of inflammatory cells. These anti-inflammatory actions of macrolides may be beneficial in attenuating inflammatory processes involved in bacterial sepsis. Therefore, we investigated the ability of azithromycin to attenuate the deleterious effects of lipopolysaccharide (LPS), in three different LPS-induced inflammatory models. Our results show that azithromycin (10 and 100 mg/kg) significantly attenuated the in...

Intestinal lactase-phlorizin hydrolase, sucrase-isomaltase and maltase-glucoamylase are brush-border hydrolases important for the digestion of carbohydrates. Alkaline phosphatase is a metalloenzyme generally used as a marker of epithelial cell maturation. The expression of these enzymes during postnatal development has been characterized, but the effect of ageing on disaccharidase activity is still poorly understood. Our aim was to determine the influence of ageing on lactase, sucrase, maltase a...

The anti-inflammatory potential of azithromycin in chronic obstructive pulmonary disease (COPD) patients was explored following a 17 standard oral dosing regimen. Patients with moderate and severe COPD were treated with azithromycin (500 mg, n =16) or placebo (n =8) 18 once daily for 3 days in a randomized, double blind design, to compare effects on inflammation markers with those seen in a previous study 19 in healthy volunteers. A battery of tests was made on serum, blood neutrophils and sputu...

Macrolide antibiotics have an outstanding ability to concentrate within host cells, particularly phagocytes. In the study described in this paper five different macrolide antibiotics were compared regarding the uptake and release kinetics in human peripheral blood polymorphonuclear neutrophils (PMNs) and three different cell lines, two phagocytic cell lines (RAW 264.7 and THP-1) and an epithelial cell line (MDCK). Based on the results obtained, the substances tested could be clustered into diffe...
