Background— Serum- and glucocorticoid-responsive kinase-1 (SGK1), a serine-threonine kinase that is highly expressed in the heart, has been previously reported to regulate sodium channels. Because SGK1 is a PI 3-kinase–dependent kinase with structural homology to Akt, we examined its regulation in the heart and its effects on cardiomyocyte (CM) apoptosis and hypertrophy in vitro. Methods and Results— Rats were subjected to aortic banding, and expression of total and phosphorylated SGK1 was exami...
Effective cardioprotection in clinical settings requires not only cardiomyocyte survival but also preservation of fonction. Multiple growth factors protect the heart from ischemic and other injury. While the downstream signaling pathways of these cardioprotective factors are complex, activation of phosphoinositide 3–kinase (PI 3–kinase) and its downstream effector, the serine-threonine kinase Akt (or Protein Kinase B), is a common feature in many cases. Genetic manipulations in cardiomyocytes bo...

Abstract Chemokines such as the monocyte chemol attractant protein-1 (MCP-1) convert monocyte rolling to firm arrest under physiological flow conditions via integrin activation and simultaneously activate phosphoinositide 3-kinase (PI3K). Here we used adenoviral gene transfer and biochemical inhibitors to manipulate PI3K-dependent pathways in human monocytes. In in vitro lipid kinase assays from purified human monocytes, we showed that MCP-1 activates the “classical” PI3Kα pathway and not PI3Kγ,...
