Abstract Cronobacter sakazakii is a common foodborne pathogen that can tolerate various stress conditions. Acidic environment is a common stress condition encountered by bacteria in food processing and gastrointestinal digestion, including both inorganic and organic acids. In order to elucidate the Acid Tolerance Response (ATR) of C. sakazakii, we performed high-throughput RNA-seq to compare gene expression under hydrochloric acid and citric acid stresses. In this study, 107 differentially expre...

Abstract Response mechanism of Cronobacter sakazakii under stress of combination of citral and carvacrol was investigated by transcriptome sequencing. Twenty-five significantly differentially expressed genes were identified to be closely related to the ability of C. sakazakii tolerating the stress after analysis, mainly involving glycerol metabolism; citrate metabolism; formate metabolism; ribosome function and transmembrane transport system. Combination of carvacrol and citral could cause dissi...

Gastrodin, a bioactive compound extracted from the plant source of Gastrodia elata Blume, has a wide range of therapeutic effects on central nervous system (CNS) diseases, but suffers from poor brain permeability and short half-life in plasma. In this study, fatty acid esters of gastrodin were successfully synthesized by a whole cell-based biocatalytic method. Aspergillus oryzae cells showed different catalytic activities in the organic solvent systems tested. Tetrahydrofuran was confirmed as th...

Puerarin, an important isoflavonoid from the edible root of Pueraria lobata, shows multiple bioactivities but suffers from low bioavailability. In this study, a new whole-cell catalytic method for acylation modification of puerarin was developed. Among the 12 strains tested, Aspergillus oryzae showed the highest catalytic activity and selectively catalyzed acylation of puerarin at the 6″-hydroxyl group. The organic solvents used significantly influenced the catalytic efficiency of the cells. In ...

Abstract The adaptability of Escherichia coli O157:H7 to different environmental stresses, including prolonged cold stress, poses a threat to food safety. In order to elucidate the prolonged cold response in E. coli O157:H7, genes related to prolonged coldresistance and their expression changes in E. coli O157:H7 ATCC35150 were screened and identified by analyzing their transcriptomedifferences between normal (37 °C) and low temperature storage (4 °C or −20 °C, 10 d), using RNA-sequencing. Accor...

Abstract Salmonella is a major foodborne pathogen throughout the world and able to resist extreme acid stress such as stomach gastric juice after mild acid adaptation, which makes it a potential hazard in food industry. We conducted a comparative transcriptomic analysis by high-throughout RNA-seq to elucidate the Acid Tolerance Response (ATR) in Salmonella Enteritidis. Among these 554 differentially expressed genes (DEGs) identified in this study, the expression of 280 DEGs were up-regulated and...

Acylation modification of phenol glycosides is currently of great interest due to the improved bioavailability and multiple functions. In this work, mono- or diesters of arbutin, an important phenol glycoside derivative, can be controllably synthesized by using whole-cell biocatalytic systems. Among fourteen microbial strains selected, Candida parapsilosis cells showed the best catalytic activity and high organic solvent tolerance. Compared with the best pure solvent tetrahydrofuran, the use of ...

Abstract Cronobacter is a group of opportunistic pathogen responsible for necrotizing enterocolitis, meningitis and septicaemia especially to infant and neonate, with high lethality ranging in 40%–80%. Some of them are able to survive in infant milk formula and still possesses capability of pathogenicity and virulence, biofilm formation, and high resistance to elevated osmotic pressure, low pH, heat, oxidation and desiccation. The genus Cronobacter consists of a diverse group of Gram-negative ba...

A combination method of real-time PCR and high-resolution melting (HRM) analysis for the rapid detection and specific classification of six pathogenic Cronobacter species based on gene cgcA was developed. HRM profiles with distinct Tm (melting temperature) peaks were consistently obtained with each species, represented by a single peak ranging from 86.02 to 86.80 °C. The detection limit ranged from 0.1 to 1 pg per reaction. Desiccated Cronobacter sakazakii and Cronobacter muytjensii of 2 cfu/25 ...
